Having actually experienced both sides of the fence... first of all, would we be talking gender (as in an individual's outside appearance) or sex? Sexologists tend to discern the two.
First thing I would do upon finishing my sexual transition? Crack open a bottle of hard liqueur and party. Assuming we are talking a return to square one in terms of my gender, likely do something horrible.
That being said, it comes down to a matter of personal comfort. A person's brain is sexed in accordance with one sex or the other, so there is a sex each will be more comfortable with -- the way you 'know' is when you are not. The latter occurrence can be frightening, because so much of our identity is based around gender and sex. It is hard to realise the massive extent, until you have tried changing your own.
To be completely honest: superficially speaking, living as a man or a woman is not too much different.
The biggest contrasts are a matter of socialisation (id est, times when you are with other men or women) and that of what mainlining the hormones on either side will do to you.

I think OP is strictly referring to physical sex. I know sex and gender are distinctly different to those who are very involved with the differences (and by many written definitions), but the terms are also used interchangeably by many.

That was what I figured, too. Tend to see these sorts of topics pop up with relative frequency, so I went into detail to 'share the goods'. Sex always pertains to biology, and gender covers most everything else.
