NPC aren't something that there are 'a few of' to consider - this is a city of 400,000 people. If we hypothetically managed to gather even as many as two hundred player characters, all together they would still drops in the ocean.
You should be aware of the demographics at all times and imagine throngs of life around your characters at nearly all places and all times of the day. You are expected to utilize them as much as possible! The only major NPC that are restricted to staff are generally those wielding great power such as The Queen, the Captain of The Guard, etc.

The following listings are for reference as to what is going on in the city and give you an idea of potential jobs and skills a character might have. It's by no means exhaustive and the following fantastic list (which organization has been borrowed from) has additional ideas:

It should never be forgotten that this is a very large trade city and as such there are multiples of everything - no matter what you specialize in or what kind of business you run, odds are excellent that you've got competition.
The exceptions are certain positions of which there is only one position open, including:
Sovereign Ruler
Royal Adviser
Captain of the Guard / Military Head
Captains of Guard(s)
Province Governors
Shipyard Captain
Crypt Keeper
Head Architect
Head Surgeon / Healer
Master of Colosseum
Stable Head
Merchant Guild Council
Head of Slave Market
Government
